蛋白酶K是一種枯草蛋白酶類的高活性蛋白酶,用于生物樣品中蛋白質的一般降解。從林伯氏白色念球菌(tritirachium album limber)中純化得到。蛋白酶K是一種切割活性較廣的絲氨酸蛋白酶。它切割脂族氨基酸和芳香族氨基酸的羧基端肽鍵。此酶經純化去除了RNA酶和DNA酶活性。由于蛋白酶K在尿素和SDS中穩定,還具有降解天然蛋白質的能力。

蛋白酶K貯存
干粉狀態可在-20℃條件下長期儲存。溶解后分裝為適當體積,2-8℃短期儲存,-20℃長期儲存。正確的貯存條件下,干粉狀態的有效期可達一年。液體狀態在-20℃有效期為半年。打開包裝使用后,如果在2~8-20℃環境下放置超過一周時間,建議過濾除菌或增加穩定劑如0.5%疊氮化鈉,防止微生物污染。
貯存和稀釋緩沖液:50mM Tris-HCl緩沖液,2mM CaCl2 (pH 8.0)。
